Martina Navratilova was the defending champion, but was lost by Jennifer Capriati in the quarterfinals.

Steffi Graf defeated Gabriela Sabatini 6–4, 3–6, 8–6 in the final to win the Ladies' Singles title at the 1991 Wimbledon Championships. After splitting the first two sets, Sabatini served twice for the title, and was two points away from her second Grand Slam win before Graf rallied. With the victory, Graf captured her third Wimbledon Singles Championship, while it proved to be Sabatini's third and final appearance in a Grand Slam final.

The seeded players are listed below. Steffi Graf is the champion; others show the round in which they were eliminated.

Monica Seles, the World no. 1 and supposed to be the first seed, who withdrew before the tournament draw was made. The seeds numbered 2-15 all rose one place and a new no.16 was added.
