South Wayne is a village in Lafayette County, Wisconsin, United States. The population was 489 at the 2010 census.

According to the United States Census Bureau, the village has a total area of 0.79 square miles (2.05 km2), all of it land.
As of the census of 2010, there were 489 people, 206 households, and 121 families residing in the village. The population density was 619.0 inhabitants per square mile (239.0/km2). There were 220 housing units at an average density of 278.5 per square mile (107.5/km2). The racial makeup of the village was 99.2% White, 0.6% African American, and 0.2% from other races. Hispanic or Latino of any race were 0.4% of the population.
There were 206 households of which 31.6% had children under the age of 18 living with them, 45.1% were married couples living together, 6.8% had a female householder with no husband present, 6.8% had a male householder with no wife present, and 41.3% were non-families. 35.9% of all households were made up of individuals and 17.5% had someone living alone who was 65 years of age or older. The average household size was 2.37 and the average family size was 3.07.
