Hopkinton is a city in Delaware County, Iowa, United States. The population was 628 at the 2010 census.

Hopkinton was laid out in 1850, and it was incorporated in 1874.

The city is the former home of Lenox College, a small school that closed in 1944. The old campus is still maintained as Delaware County Historical Museum Complex.

Hopkinton is located at 42°20′38″N 91°14′55″W / 42.34389°N 91.24861°W / 42.34389; -91.24861 (42.3438857, -91.2484799), near the Maquoketa River. According to the United States Census Bureau, the city has a total area of 0.62 square miles (1.61 km2), all of it land.

As of the census of 2010, there were 628 people, 266 households, and 185 families residing in the city. The population density was 1,012.9 inhabitants per square mile (391.1/km2). There were 294 housing units at an average density of 474.2 per square mile (183.1/km2). The racial makeup of the city was 99.0% White, 0.3% from other races, and 0.6% from two or more races. Hispanic or Latino of any race were 0.8% of the population.

There were 266 households of which 30.8% had children under the age of 18 living with them, 51.5% were married couples living together, 12.0% had a female householder with no husband present, 6.0% had a male householder with no wife present, and 30.5% were non-families. 25.9% of all households were made up of individuals and 13.6% had someone living alone who was 65 years of age or older. The average household size was 2.36 and the average family size was 2.76.
