The following is a list of characters who first appeared in the BBC soap opera Doctors in 2012, by order of first appearance. All characters are introduced by the series' producer Peter Eryl Lloyd. The first main character to be introduced was Mandy Marquez, a love interest for Freya Wilson and Akono Mezu, a student who moves in with Mrs Tembe. February saw the introductions of Malcolm and Paula Malone and Franklyn Ward who arrive for the wedding of Cherry and Jimmi Clay. February also saw the birth of Joe Carmichael, the son of Daniel Granger and Zara Carmichael. Martin Millar arrived in April, part of the storyline to coincide with the departure of Julia Parsons, Howard Bellamy arrived as her replacement in May, and Jas Khella, a friend of Kevin Tyler's, arrived at the Mill in August. Grace Jacobs arrived as Jas's colleague in September, as did Kingsley Apollo, a love interest for Imogen Hollins. Harrison Kellor returned in September, along with the introductions of Helen Callaway and Alex Redmond, and Emma Reid arrived in October, a replacement for Elaine Cassidy, who departed due to Harrison's return. Valerie Pitman and Emma's son Chris also arrived in October, the latter a replacement for Cherry.

Mandy Marquez played by Danielle Henry, made her first appearance on 10 February 2012. Mandy is a love interest for Freya Wilson (Lu Corfield). Mandy is introduced as a bridesmaid to Cherry Malone (Sophie Abelson). Freya later begins to develop feelings for Mandy, and the two meet up. Mandy, at first, is not sure whether she likes Freya as she is bisexual, though later begins to like her, and the two go out for dinner. The following week, Freya takes part in a football game, and Mandy comes to watch. When Mandy gets there, the team is a player short, so Mandy is asked to play. They win, and Mandy asks Freya back to her house.
