Maria Sharapova was the defending champion, but withdrew from the tournament, due to a recurring shoulder injury which also ruled her out of the 2008 US Open.

Serena Williams won in the final 6–0, 6–3, against Dinara Safina in just under one hour. It was Williams' fourth career Australian Open singles title, her tenth Grand Slam title and Safina's second career Grand Slam final loss. This led to Williams returning to the No. 1 ranking for the first time since October 2008.
