Andre Agassi defeated defending champion Pete Sampras 4–6, 6–1, 7–6(8–6), 6–4 in the final to win the Men's Singles tennis title at the 1995 Australian Open.

Agassi competed in the Australian Open this season for the first time in his career. He would go on to win three more in 2000, 2001, and 2003.

A memorable moment occurred in the quarterfinal match between Sampras and Jim Courier. Sampras had fought back from a 2-sets down deficit to tie it at 2-2. Shortly after winning the first game of the fifth set, Sampras was overcome with grief over his then-coach, Tim Gullikson. Gullikson had suffered several strokes while guiding Sampras during the tournament and was rushed to a hospital in Chicago shortly before the match. Sampras retired to his chair and broke down in tears. When he returned to the game, he found it difficult to continue and Courier offered to finish the match the next day. Sampras declined and was able to close out the match. Gullikson was diagnosed with inoperable brain cancer and would pass away almost a year later.
